We're talking about Trigun here right?  Vash only kills one character in all 26 episodes on "purpose"... which is Degato.  The first time he used the Angel Arm Cannon and annihalited that town with the seasonal name (Augusta maybe?) was only when Nives forced him to using his powers.

Rodimus did in TF:TM and Optimus Supreme did in Energon (though Unicron came back later)

 

Also in Energon, they used the Energon shield to kill Demolisher (though he came back too! but almost everyone comes back again in a new body anyway). Plus they destroyed alot of Terrorcons.
